The film opens with Spider-Man captured by Colonel John Jameson and his son J. Jonah Jameson. The entire Jameson family watches as Spider-Man's alter ego Peter Parker is forced to explain why he fights crime across New York City, while also trying to protect their heir apparent J. Jonah until the Colonel calls it off due to lack of evidence for his claims about Spider-Man being a menace in New York City. After reliving the events of his battle with Eddie Brock, Spider-Man manages to free himself and recovers from the apparent death that he had at the end of Spider-Man 2. He then takes on Captain Stacy's team and easily defeats them for breaking into Oscorp and stealing the formula for his synthetic webbing.As he moves towards his office, Spider-Man is attacked by a group of gangsters called "The Enforcers" who are attempting to steal the formulas for his webbing. He quickly defeats them but is unable to prevent one of them from shooting him with a gun loaded with tranquilizer darts. Spider-Man then passes out and is taken by the Enforcers to Dr. Ashley Kafka who assures them that she will be able to 'cure' Spider-Man.The next day, Peter Parker wakes up in a makeshift prison cell at the top of a tower surrounded by armed guards and security cameras. Spider-Man manages to make his way out of his cell and discover that he is on an island owned by Oscorp owned by Dr Ashley Kafka. He also discovers he has been injected with a new synthetic webbing formula which cannot be removed, leaving him with only one functioning arm for the duration of his stay on the island. Dr Kafka sends armed guards after Spider-Man once he leaves his cell. As the guards are forced to retreat due to not being able to reach their weapons, Spider-Man uses webheads to defeat them. He also discovers that the synthetic webbing formula is used by J. Jonah Jameson for his satellite show "The New York Post". Spider-Man decides to call his old friend Harry Osborn, who is now the owner of Oscorp, and asks him if he can find a way for him to leave the island without being recaptured by heavily armed guards. Harry refuses to help, as Oscorp now owns the island, and as a result Spider-Man is forced to use his normal webbing to escape from the guards. Harry later has a change of heart and decides to help Spider-Man however he can. He reveals that Dr Kafka is planning on using his new formula on himself and upon discovering this information Colonel Jameson attacks Harry for violating Oscorp's 'no harm' policy. Harry survives and escapes with Spider-Man but is then captured by J. Jonah Jameson's guards. While escaping from Jameson's helicopter, Spider-Man makes it into the air where he manages to trash it before falling out of the helicopter onto the city below.
